21 Is there a preferred signal peptide to use with pCHO 1.0 and should I optimize my sequence? • We cannot give any specific recommendations regarding which signal peptides to use. However, we highly recommend that any sequence, including genes and signal peptides, be at least evaluated for optimization via GeneArt ® services in order to avoid, among other things, cryptic splice sites, and to improve CHO-specific codon usage (GeneArt ® gene optimization services include searches for other potential issues as well). We know that cryptic splice sites can severely compromise titer. Users can decide whether or not to go through with synthesis of the optimized gene after they get the GeneArt ® services report. • Unless it is optimized, avoid using the following signal peptide, derived from a murine Ig heavy chain locus, in the pCHO vector (even if it's been successful for secretion in another vector/cell line): — ATG GGA TGG TCA TGT ATC ATC CTT TTT CTA GTA GCA ACT GCA ACC GGT GTA CAT TCA For more detailed information, please see the "Creating Expression Plasmids for the Freedom ® CHO-S ® Kit" section of the User Guide.
