Q: A: 34 Life Technologies ™ | Cell Clumping During Freedom ® CHO-S ® Workflow Measure viable cell density and viability AFTER straining cells. Don't overgrow cells during pre-transfection passage. Keep viable cell density under 2 x 10 6 viable cells/mL. Don't use Anti-Clumping Agent prior to transfection. How do I deal with CHO-S ® cell clumping? Clumping with CHO-S ® cells is to be expected. There are three ways to minimize cell clumping: 1. Remove clumps with a cell strainer beginning at passage 1 post-thaw. 2. Avoid cell clumps in both sampling and passage of CHO-S ® cells. 3. Do not allow CHO-S ® cell densities to exceed 2 x 10 6 viable cells/mL pre-transfection. Do not, however, use Anti-Clumping Agent prior to transfection. Later, inclusion of Anti-Clumping Agent is critical to the success of selection. Cell Clumping During Freedom ® CHO-S ® Workflow Don't: Do:
