38 Life Technologies ™ | Transfection with FreeStyle™ MAX Reagent Passage CHO-S ® cells at least 5x prior to transfection. Only transfect cells that are ≥95% viable. Check carefully for DNA precipitation upon addition of diluted FreeStyle ™ MAX Reagent to diluted DNA (Figure 5). Use a vessel large enough for proper mixing of the final transfection mixture (e.g., a 50 mL conical centrifuge tube). Add diluted FreeStyle ™ MAX Reagent to diluted DNA by first immersing the tip and expelling the solution slowly. Allow complexes to form for at least 10 minutes before adding mixture to cells. Add the DNA:FreeStyle ™ MAX Reagent complex dropwise into a 125 mL shake flask containing cells while slowly swirling the flask. Don't use Anti-Clumping Agent in transfection or in passage prior to transfection. On the day of transfection, don't centrifuge the cells if the cell count is <1 x 10 6 cells/mL.
