Q: A: 57 How long does it take to select for a stable pool? How low does viability drop? Phase I selection typically takes 14–18 days, and Phase II selection 7–14 days, for a total of 21–32 days. Viability in Phase I can drop below 10%; in Phase II, it usually does not drop below 40%. In this example (see Figure 13), it took a total of 22–32 days to complete both selection phases. The no-DNA control will drop below 10% beyond the first week in selection, and will help to distinguish true recovery during selection.
