59 If you cannot clone from the stable pool cell train upon recovery, work from a frozen vial as follows: thaw the stable pools to be cloned into CD FortiCHO ™ Medium supplemented with glutamine (Anti-Clumping Agent is optional; selection pressure is not recommended) and seed for limited dilution cloning 2–3 days later (without additional pool passage). Pools should be ≥95% viable before proceeding with limited dilution cloning. While all steps preceding selection, including pre-transfection passage and transfection of CHO-S ® cells, have been done using 130–150 rpm, we have found more consistent results in selection using 150 rpm. Whatever rpm setting is used during selection, we recommend keeping that setting for all subsequent shake flask pool and clone handling steps, including productivity assessments.
