Tip: Tip: 32 Life Technologies ™ | Cell Clumping During Freedom ® CHO-S ® Workflow Cell clumping creates different issues depending on the stage of the workflow. We have successfully used a cell strainer at every cell passage throughout the workflow, both pre-transfection and during selection, without negative effects. Thus, if in doubt, strain the cells. However, it is less labor-intensive to use the cell strainer on an as-needed basis (see video). Clumping on transfection day: Cell clumping decreases transfection efficiency, so cell straining is highly recommended on the day of transfection to ensure that every cell is sufficiently exposed to transfection reagents. Clumping during selection: Cell clumping decreases pool productivity by trapping high-producing cells in clumps that do not get passaged, so cell straining is critical during selection to distribute high-producing cells throughout the culture prior to passage.
